Phosphoramidites, first described in 1981, are modified nucleosides, which allow for the addition of new base sequences to the DNA strand in an extremely simple and unusually effective cyclic reaction. Before phosphoramidite-based synthesis, the length, quality, and yield of DNA synthesis were limited, due to the highly unstable synthesis reaction intermediates. Phosphoramidites contain the best protecting group, which makes the chemical properties of phosphoramidite different from other methods.
